A toughness based deformation limit for X- and K-joints under brace axial tension

Abstract: This study reports a deformation limit for the initiation of ductile fracture failure in fatigue-cracked circular hollow section (CHS) X- and K-joints subjected to brace axial tension. The proposed approach sets the deformation limit as the numerically computed crack driving force in a fatigue crack at the hot-spot location in the tubular joint reaches the material fracture toughness measured from standard fracture specimens. The calibration of the numerical procedure predicates on reported numerical computations on the crack driving force and previously published verification study against large-scale CHS X-joints with fatigue generated surface cracks. The development of the deformation limit includes a normalization procedure, which covers a wide range of the geometric parameters and material toughness levels. The lower-bound deformation limits thus developed follow a linear relationship with respect to the crack-depth ratio for both X- and K-joints. Comparison of the predicated deformation limit against experimental on cracked tubular X- and K-joints demonstrates the conservative nature of the proposed deformation limit. The proposed deformation limit, when extrapolated to a zero crack depth, provides an estimate on the deformation limits for intact X- and K-joints under brace axial loads.

Abstract: Cullin-RING E3 ubiquitin ligase (CRL)-4 is a member of the large CRL family in eukaryotes. It plays important roles in a wide range of cellular processes, organismal development, and physiological and pathological conditions. DDB1- and CUL4-associated factor 8 (DCAF8) is a WD40 repeat-containing protein, which serves as a substrate receptor for CRL4. The physiological role of DCAF8 is unknown. In this study, we constructed knockout mice. Homozygous mice were viable with no noticeable abnormalities. However, the fertility of deficient male mice was markedly impaired, consistent with the high expression of DCAF8 in adult mouse testis. Sperm movement characteristics, including progressive motility, path velocity, progressive velocity, and track speed, were significantly lower in knockout mice than in wild-type (WT) mice. However, the total motility was similar between WT and knockout sperm. More than 40% of spermatids in knockout mice showed pronounced morphological abnormalities with typical bent head malformation. The acrosome and nucleus of knockout sperm looked similar to those of WT sperm. tests showed that the fertilization rate of knockout mice was significantly reduced. The results demonstrated that DCAF8 plays a critical role in spermatogenesis, and DCAF8 is a key component of CRL4 function in the reproductive system.

Abstract:

S100A9, a calcium-binding protein, participates in the inflammatory process and development of various tumors, thus attracting much attention in the field of cancer biology. This study aimed to investigate the regulatory mechanism of and its function involvement in APL. We used real-time quantitative PCR to determine whether PML/RARα affects the expression of in NB4 and PR9 cells upon ATRA treatment. ChIP-based PCR and dual-luciferase reporter assay system were used to detect how PML/RARα and PU.1 regulate promoter activity. CCK-8 assay and flow cytometry were employed to observe the viability and apoptosis of NB4 cells when was overexpressed. Results showed that was an ATRA-responsive gene, and PML/RARα was necessary for the ATRA-induced expression of in APL cells. In addition, PU.1 could bind to the promoter of , especially when treated with ATRA in NB4 cells, and promote its activity. More importantly, overexpression of induced the apoptosis of NB4 cells and inhibited cell growth. Collectively, our data indicated that PML/RARα and PU.1 were necessary for the ATRA-induced expression of in APL cells. Furthermore, promoted apoptosis in APL cells and affected cell growth.

Abstract: In this paper, the research framework for specific structure crystallization modeling has been proposed in which four steps are required in order to investigate the rigorous crystallization modeling by thermodynamics. The first is the activity coefficient model of the solution, the second is Solid-Liquid equilibrium, the third and fourth are the dissolution and crystallization kinetics modeling, respectively. Our investigations show that the mechanisms of complex structure formation and microphase transition can be analyzed by combining the dissolution and crystallization kinetics modeling. Moreover, the formation mechanism of the porous KCl has been analyzed, which may provide a reference for the porous structure formation in the advanced material synthesis.
